AB OBJECTIVE: HIV-I Rev protein and HTLV-I Rex protein possess their own
conserved leucine-rich domains which are thought to interact with
cellular factors. Our aim has been to isolate and characterize these
co-factors via biochemical approaches. METHODS: Oligopeptides
corresponding to the motifs of Rev (EPVPLQLPPLERLTLDCNED) and Rex
(MDALSAQLYSSLSDSPP) were chemically synthesized and coupled to Sepharose
4B resin by chemical cross-linking. The affinity resin was used to
isolate cellular factors and to carry out various kinds of protein
binding analysis. RESULTS: By affinity-chromatography of the whole cell
extract from MOLT-4 cells with conjugated Rev-peptide, we partially
purified a cellular protein of approximately 18KDa. Interestingly, the
purified protein was able to interact not only with the Rev-derived
peptide but also with the Rex specific peptide. DISCUSSION AND
CONCLUSIONS: It was reported that eukaryotic initiation factor 5A
(eIF-5A) is a cellular target for the leucine-rich domain of Rev, but
the purified protein was no longer active in binding assays (Ruhl et
al., J. Cell Biol. 123:1309, 1993). Studies are ongoing to characterize
and determine the exact nature of the binding protein we have isolated
and found to interact with both the Rev and Rex peptides.
